空军一号是对搭载美国总统的任何美国空军飞机的称呼，目前美国总统最常用的是两架VC-25A飞机，尾号28000和SAM 29000。两架飞机是改装过的波音747-2G4B民航机。在28000和SAM 29000于1990年开始服役之前，空军一号是两架波音707-320B飞机，尾号26000和27000，这两架飞机从1958年起就开始服役了。
空军一号主要改装了飞机的内部装璜和设施，以更适合总统在空中行驶其职责，飞机上有休息区、办公区、两个厨房、一个手术台、药房、通讯系统、电话和电视机等。此外还特别为总统的家人、白宫员工和媒体记者保留了空间。在诸如核战争等紧急状态中，空军一号也可以成为一个军事命令中心。此外还加入了空中加油的设备以及反地对空系统。
空军一号的所有飞行都属于空军的军事行动，由位于马里兰州的安德鲁空军基地的总统飞行组负责。从白宫到安德鲁空军基地总统一般先要搭乘美国海军陆战队的专用直升机海军陆战队一号。同样的，搭承总统的陆军交通工具被称为陆军一号，搭承总统的海军交通工具被称为海军一号，而搭载总统的民航飞机则被称为执行一号（Executive One）。
将总统的飞机称为“空军一号”是在艾森豪威尔总统时代开始的，主要是为了安全原因。当年有一个拥有与总统飞机同样代号的民航飞机正好与总统专机一起进入同一个空域，从此之后总统的飞机就有了一个特殊的名字，以和其他所有航班区别。
两架总统专机只有当总统在飞机上时才被称为空军一号。当1974年尼克松总统辞职，然后搭乘总统专机离开安德鲁空军基地时，飞机的代码被改成了飞机的尾号，只有当继任的福特总统正式接任后，才恢复了空军一号的代号。
