The Golgi Apparatus: The First 100 Years traces the first 100 years of Golgi apparatus discovery from the first published accounts from Pavia, Italy in 1898 to the Centenary Celebration in Pavia, Italy in 1998 and into the decade beyond. It is not intended, however, to be a comprehensive survey but rather to present the perspectives of the authors to summarize their contributions over the past 50 years in parallel with the modern era of Golgi apparatus discovery initiated in 1954 and made possible by the advent of the electron microscope. Included are methods of cell fractionation and biochemical analysis leading up to the present where efforts focus heavily on molecular biology. Topics where the authors and their colleagues have made substantial and/or pioneering contributions are emphasized including Golgi apparatus morphology and structural organization and function (especially in plants), the existence and importance of cisternal tubules, development of methods of plant and animal Golgi apparatus isolation and subfractionation, biochemical analyses of highly purified plant and animal Golgi apparatus fractions in comparison to equally highly pu- fied reference fractions.

高分子物理最经典的教材之一。难是真的难,但读明白了30%依然受益匪浅。随着阅读的进程大概也感受到了一点为什么这么多做生物物理/软物质的人如苍蝇嗅到鲜血般一窝蜂地涌向“相分离”领域。物理真的是可以用最简单的概念+空间和时间尺度的分离来解释无比复杂的观测。溶剂,单体浓度,聚合度比例之间的调和,熵和相互作用(力和内能)之间的调和,仅仅是这样就可以创造出无比复杂的软物质世界。——回到自己感兴趣的部分:想从复杂的公式和推导中获得一些对生命体系有意义的直感。从这个角度讲目的已经达到了。

花了半年终于读完了。第一部分很简单的一点分子生物学;之后是以病原为核心的微生物学——这部分读起来最开心,非常有意思;最后一部分比较偏临床生理,讲得不系统而且一大堆让人头疼的名词...一本读下来感觉长了很多姿势,非常感叹人类的渺小和各种生命体的复杂性。

嗜极生物的百科全书。读了半年多终于读完了,受益良多。针对每一种极端环境中的生物基本都是从环境的物种多样性开始,到基因组学,再到通路、分子和生物物理。比较有趣的是,偏离温和条件的两种极端采取的手段往往并非完全相反。比如对付低温的分子机理并非是把对付高温的机理反其道行之。读完感觉PV=nRT真的是个伟大的方程,囊括了一般能想到的环境里几乎一切的物质和状态:嗜高低温的核心是大分子结构稳定性和动态在熵焓层面的博弈,到了嗜盐卤开始出现了平衡静电相互作用,再到嗜酸碱部分质子浓度高低导致重点变成了化学势、电位和代谢,然后是研究较少的高压对密度和体积的讨论。最后一章还有点哲学思考,反思了一下过分的无孔不入的“达尔文主义”对思考生命起源的禁锢,非常精彩。

上次读系统发生学(为什么Phylogenetics的中文翻译这么奇怪)的书可能还是4-5年前。这本书感觉写的特别好,从基本概念的定义开始捋,最后引出重量级的中性演化对自然选择大战。整体来说对基因漂变和近中性演化的部分理解更好了。不过限于自己低下的概统基础,一到不太直观的地方还是会卡壳...最后一章的实例也比较有意思,当时还是H5N1禽流感肆虐。恐怕再过10年会单独出一章分析新冠病毒的流行和溯源了吧...

胞内膜泡输运的经典参考书。虽然有很多部分比较旧了,但是知识梳理特别清晰,非常合适捋一捋细胞生物学的基础。还是有一种近15年分子细胞领域进入了参考书真空期的感觉,很多新的知识没有人愿意花时间进行系统编纂。可能也是一种短平快评价体系的客观反映。这本书让我眼前一亮的地方是很多章最后会有一小段介绍在成书期间这一章里面未解的知识有什么新的动态,真的很棒!另外就是觉得做信号转导的人写出来的东西怎么这么无聊...完全没有放眼于整个细胞生命过程的感觉,就盯着自己熟悉的那些个通路和基因缩写一阵天花乱坠......

从演化生物学的视角看每个细胞器。是一本编得很不错的细胞生物学和演化生物学的交叉参考书。前三章更是非常难得地从宏观的角度去分析真核细胞及其内膜系统可能的起源。当下的系统发生学和分子演化太蛋白质中心了,这本书虽然也不例外,不过已经做得足够好了。唯一的遗憾是成书年代太早,2007年,很多关键复合物的结构和机理都还没有阐明。要是现在能再版就好了——可现在几乎也没有人有这个耐心去做系统性的编纂了。

从实验室薅到这本书两年多尝试多次之后,终于能把这本书从头到尾读下来了。收获巨大脑子也巨大。可能自己本科背生化代谢的时候很难想到有一天要集中面对一整本的三字母+数字的基因名。因为实际的代谢网络太复杂,所以虽然系统,但细节仍然无法一一讲清。最后一章有大量的图号对不上的情况有点难受。酵母的系统就是太versatile了,各个通路都充满着冗余和旁路。不禁再一次佩服解析代谢通路的遗传和生化学家们的智慧(我再看看现在这堆只知道堆omics的所谓“大”科学家们)。本书经典结语是一句平实有力的废话:“fooling with metabolism has many effects.”

It is a truth universally acknowledged, that a single man in possession of a good fortune, must be in want of a wife."Next to the exhortation at the beginning of Moby-Dick, "Call me Ishmael," the first sentence of Jane Austen's Pride and Prejudice must be among the most quoted in literature. And certainly what Melville did for whaling Austen does for marriage——tracing the intricacies (not to mention the economics) of 19th-century British mating rituals with a sure hand and an unblinking eye. As usual, Austen trains her sights on a country village and a few families——in this case, the Bennets, the Philips, and the Lucases. Into their midst comes Mr. Bingley, a single man of good fortune, and his friend, Mr. Darcy, who is even richer. Mrs. Bennet, who married above her station, sees their arrival as an opportunity to marry off at least one of her five daughters. Bingley is complaisant and easily charmed by the eldest Bennet girl, Jane; Darcy, however, is harder to please. Put off by Mrs. Bennet's vulgarity and the untoward behavior of the three younger daughters, he is unable to see the true worth of the older girls, Jane and Elizabeth. His excessive pride offends Lizzy, who is more than willing to believe the worst that other people have to say of him; when George Wickham, a soldier stationed in the village, does indeed have a discreditable tale to tell, his words fall on fertile ground.
Having set up the central misunderstanding of the novel, Austen then brings in her cast of fascinating secondary characters: Mr. Collins, the sycophantic clergyman who aspires to Lizzy's hand but settles for her best friend, Charlotte, instead; Lady Catherine de Bourgh, Mr. Darcy's insufferably snobbish aunt; and the Gardiners, Jane and Elizabeth's low-born but noble-hearted aunt and uncle. Some of Austen's best comedy comes from mixing and matching these representatives of different classes and economic strata, demonstrating the hypocrisy at the heart of so many social interactions. And though the novel is rife with romantic misunderstandings, rejected proposals, disastrous elopements, and a requisite happy ending for those who deserve one, Austen never gets so carried away with the romance that she loses sight of the hard economic realities of 19th-century matrimonial maneuvering. Good marriages for penniless girls such as the Bennets are hard to come by, and even Lizzy, who comes to sincerely value Mr. Darcy, remarks when asked when she first began to love him: "It has been coming on so gradually, that I hardly know when it began. But I believe I must date it from my first seeing his beautiful grounds at Pemberley." She may be joking, but there's more than a little truth to her sentiment, as well. Jane Austen considered Elizabeth Bennet "as delightful a creature as ever appeared in print". Readers of Pride and Prejudice would be hard-pressed to disagree. ——Alix Wilber

也许是“霸道总裁爱上我”的鼻祖www。相当慢热的作品,差点又在40%左右半途而废。一旦度过最煎熬的时候,之后几对矛盾冲突的感情描写都让人觉得既畅快淋漓又婉转细腻。就算是“有钱男人一定想要娶老婆”的时代,文学作品里也还会讴歌自由爱情的美好顺便用各种不同的角度讽刺婚姻这种制度本身;不得不又让人感慨当代究竟是怎么一个时代。

急需细胞/生理学入门基础就从图书馆薅了这本书。编的挺系统的,收获其实非常大,唯一的问题就是太老,少了20年飞速发展的知识。——我就好奇,为什么现在这个时代真正写系统性参考书的人越来越少了,出现的新书全都是专题性的...可能大家也越来越少了,比起掌握全局,售卖自己熟悉的概念,用出书来巩固自己的小圈子才是现代学术界的重中之重。

最喜欢的部分是罗素对中国、日本、西欧、美国各自文化和价值观的分析和批判。罗素的很多糟糕方面的预期都不幸成为了现实。他在当时背景下所提出的“中西结合”的最佳解决方式如今来看并没能成为现实。贯彻始末的“强大政府“并没有如愿出现,日本也并没有反思自己的扩张欲可能带来的后果,最终历史走向了他所预期的风险最高,损失最惨重的道路。而从现在这个节点去比对罗素的分析,一是感慨当代社会在精神上并没有完全规避全盘西化可能性中诸多的弊端,很多在一百年前国人精神中尚且存在的闪光和自信并没能抵抗住资本主义和工业化的侵蚀;二是他还是低估了他所总结的国人缺点中“贪婪、怯懦、无情”对中国政治走向的影响;最后则是更加凸显了毛真的改变了中国的历史。

中规中矩的一本参考书,现在来看有一半多的知识过时了。偏分子机理/物化的几章写得很好,2,4,5,10四章精读有非常大的收获。其他几章就能看出蛋白质中心的研究者们强行和脂扯关系的用力感。看里面对成像/质谱/电镜技术的描述不禁感慨15年生物物理技术的剧变。说到底”脂筏“模型不过是最可能的一种假说而已...

文字流畅简洁,情节扣人心弦,画面感极强。没有达芬奇密码那种做作的宗教炫学感,充盈的是纯粹解谜的快乐。结局最后的密码对于有一点化学常识的人来说有点配不上全篇精彩的设定——但这被尾声后余味满满的彩蛋完美补偿了!其实说这是一本悬疑小说,说它是精彩的长篇童话也不为过!
